چکیده

In the present Frontier article, we highlight recent new results on the use of platinoid complexes as therapeutic agents against Alzheimer's disease and infer the relevance of targeting monomeric soluble disordered amyloid peptides to preclude the deleterious aggregation process associated with the etiology of the disease.

The structure of intrinsically disordered peptides implicated in amyloid diseases: Insights from fully atomistic simulations Structures of the Alzheimer Amyloid-β and the IAPP peptide monomers

This book chapter reviews recent all-atom simulations that aim at characterizing the monomeric structure of intrinsically disordered peptides implicated in protein aggregating diseases. Simulations reveal that peptides that appear to be disordered based on experimental studies that make use of ensemble-averaging techniques, in fact possess local secondary structure that may play a role in initi...

Islet Amyloid Polypeptide is not a Target Antigen for CD8+ T-Cells in Type 2 Diabetes

Background: Type 2 diabetes (T2D) is a chronic metabolic disorder in which beta-cells are destroyed. The islet amyloid polypeptide (IAPP) produced by beta-cells has been reported to influence beta-cell destruction. Objective: To evaluate if IAPP can act as an autoantigen and therefore, to see if CD8 + T-cells specific for this protein might be present in T2D patients. Methods: Peripheral blood ...

NEP, ACE and Homologues: The Pathophysiology of Membrane Metalloproteases

The zinc metalloprotease, neprilysin (NEP), plays a role in the metabolism of cardiovascular, inflammatory and neuropeptides, including mitogenic peptides such as bombesin. In the cardiovascular system, NEP has a primary role in the inactivation of natriuretic peptides but also contributes to local metabolism of angiotensin, endothelins and bradykinin. Hence NEP is seen as a potential therapeut...

Methionine oxidation of amyloid peptides by peroxovanadium complexes: inhibition of fibril formation through a distinct mechanism.

Fibril formation of amyloid peptides is linked to a number of pathological states. The prion protein (PrP) and amyloid-β (Aβ) are two remarkable examples that are correlated with prion disorders and Alzheimer's disease, respectively. Metal complexes, such as those formed by platinum and ruthenium compounds, can act as inhibitors against peptide aggregation primarily through metal coordination. ...

P 102: The Study of Some Factors Which Effect on Beta-Amyloid Signaling in Neuroinflammation

Neurological inflammatory diseases are developing rapidly. Different factors involved in the pathogenesis of these diseases. In this article, we discuss some of the mechanisms are dealt with. An aberrant procedure of beta-amyloid precursor protein (BAPP) to form neurotoxic beta-amyloid peptides and an accumulated insoluble polymer of beta –amyloid (BA) that forms the senile plaque. The ab...
